A methylene-blue (MB)-labeled molecular beacon junction probe allows for a signal-on electrochemical detection of nucleic acids via target recycling using endonucleases. Electron transfer is reduced when the MB is intercalated in the stem of the molecular beacon, but then electron transfer from MB to a gold electrode is enhanced upon cleavage of the junction probe due to increased probability of MB approaching the electrode when attached to the more flexible ssDNA.  相似文献

This article focuses on the development of an innovative method, based on thermodynamic considerations and with the use of Differential Scanning Calorimetry (DSC), for the estimation of the melting enthalpy of crystalline compounds which are metastable near their melting temperature. The curves obtained, at various heating rates, are analysed in two steps. In the first step, the area of a zone generated by the melting endothermic peak is calculated following a specific method. In the second step, the melting enthalpy is extracted from this area through an enthalpy balance. This method is applied to both identified crystallographic forms, named form I and form II, respectively, of Etiracetam (UCB Pharma). The results show that the melting enthalpy of the stable form II compare well with the ones obtained using conventional methods. The curves of the metastable form I present thermal instabilities (partial solid–solid polymorphic transition and beta-recrystallization) near the form I melting peak leading to difficulties for a direct determination of the melting enthalpy by conventional methods. The proposed method is therefore very useful for the estimation of the form I melting enthalpy.  相似文献

The application of hybrid polymer precursor gel for distributed feedback (DFB) grating laser fabricated by short‐pulse laser interference has been investigated. The precursor gel was prepared by sol–gel process from siloxane‐modified methacrylate monomer. The molecular structure characterization results show the formation of inorganic networks within the precursor gel, which further undergoes the formation of organic–inorganic polymer network by photopolymerization. The laser interference was performed by using the frequency‐tripled output of nanosecond Nd–YAG laser. The fabricated DFB gratings exhibit photopumped lasing actions with high consistency between the grating periodicity (between 380 and 1030 nm) and the lasing wavelength, which appears at the wavelength of the second‐order Bragg reflection. The atomic force microcopy measurements clearly show the formation of surface relief (corrugated) structure in those gratings. The mechanism of surface relief formation is attributed to a fast photo‐induced swelling process, which is unique property belonging to this kind of hybrid polymer precursor gel. In this work, we investigate the chemistry for an aqueous acetate-triethanolamine-ammonia based YBa(2)Cu(3)O(7-δ) (YBCO) precursor system. These precursor solutions are suited for the chemical solution deposition of superconducting YBCO layers on top of single crystal SrTiO(3) or buffered NiW tapes. The development of this kind of precursor inks often involves trial-and-error experimenting and thus is very time-consuming. To reduce labwork to the minimum, the theoretical prediction of pH stability limits and the complexation behaviour of the different metal ions and complexants in the inks are very important. For this purpose, we simulated, based on literature values, the complexation behaviour of Cu(2+) in the aqueous precursor solutions as a function of pH. To validate the used model, we performed potentiometric pH titrations for solutions with similar composition and checked the correctness of fit between experiment and model. The generated complexometric results are coupled with X-band EPR spectra to further confirm the results. EPR spectra for fully prepared precursor solutions as well as for Cu(2+) reference solutions containing only one type of ligand (acetate, triethanolamine or ammonia) were investigated as a function of pH. We find that, in line with speciation simulation, only acetates are actively complexing the Cu(2+) ions at pH values below 7, while when reaching higher pH levels mainly triethanolamine complexes are formed. Over the entire pH range, no trace of free Cu(2+)or Cu(OH)(2), possibly creating precipitation during gelation and thus complicating further processing, could be found.  相似文献

The conjugation of a ribonucleic acid 16-mer with the cationic amphiphilic peptide penetratin and an anionic hyaluronan tetrasaccharide by means of Cu-free "click" chemistry is reported. The alkyne-functionalized 16-mer was prepared by automated solid-phase synthesis, using a newly developed strained cyclooctyne phosphoramidite in the final coupling. Cycloaddition of the alkyne functionalized RNA to the azide containing biomolecules led to a clean conversion into the corresponding nucleic acid conjugates.  相似文献

Three-dimensional interfacial waves developing on the free surface of falling liquid films are known to intensify heat and mass transfer. In this context, the present paper studies the effect of electrostatic as well as of thermo-capillary forces on a falling film of a dielectric liquid. Therefore, measurements of the local film thickness using a confocal chromatic imaging method were performed under isothermal and heated conditions. The experimental results show that both forces destabilize the flow. It is found, that the application of an additional electric field under heating conditions enhances the generation of rivulets, thereby reducing the overall heat transfer coefficient.  相似文献

Summary High program failure rates in the pharmaceutical industry have prompted the development of predictive software that can profile compound libraries as being ‘druglike’ (resembling existing drugs) and/or ‘leadlike’ (possessing the structural and physicochemical profile of a quality lead). In recent years, these two notions prompted pharmaceutical companies to clean up their corporate libraries of screening compounds. In order to maintain and expand the size and diversity of these corporate libraries, pharmaceutical companies still continue to add compounds to these, mainly by the acquisition of screening libraries. In this paper, we have analyzed 45 commercially available libraries, offered by suppliers of screening chemistry, for leadlikeness and diversity of the offered structures. To this end we have used a set of structural and physicochemical filters for leadlikeness that was developed in-house. These 45 supplier libraries contained a total of 5.3 million structures, of which 49% (2,592,778 structures) turned out to be unique, and only 12% (677,328 structures) were found to be both unique and leadlike. A diversity analysis revealed that big differences exist between the various offered libraries.  相似文献

Novel Bragg grating waveguide structures have been fabricated in bulk borosilicate glass through a type II photosensitivity mechanism driven by single femtosecond laser pulses. Low-loss single-mode waveguides and narrow-linewidth Bragg gratings were generated simultaneously by forming an array of refractive index voxels in a single laser scan. Laser pulse duration is shown to significantly affect the grating strength and waveguide loss. Bragg wavelengths, defined by the periodicity of laser-modified volumes, were fully controlled by the sample scan speed to provide resonances anywhere in the 1200-1620 nm telecommunication bands. Four linear Bragg filters with distinct resonant wavelengths are presented that define the first demonstration of laser writing of multiple-wavelength and cascaded Bragg grating waveguides in a single process step.  相似文献

Scalable multichannel microstructuring was demonstrated by multiplexing a focused molecular fluorine laser beam with cylindrical nanolayer microaxicons into an array of converging pseudo-nondiffracting subbeams. The axicons were fabricated by shadow-mask vapor deposition of magnesium fluoride onto substrates of identical material. Long-period surface gratings of variable pitch were generated on poly(methyl methacrylate) by varying the target position within the converging periodic focal lines.  相似文献
